As mentioned there is definitely a Hertz desk located at the Venetian (there over 20 options when you type Las Vegas as your pick up, the Venetian being one of them). For drop off location you select Las Vegas/Harry Reid airport.

   We've double booked comp rooms partially for 3 trips. Usually it's in the middle of a trip for 1-2 nights to try out other properties. We keep our Paris room for the entire trip and just take a backpack for checking into double booked room we are trying out.

   I say this because you ideally want to see Grand Canyon National Park aka South Rim. That requires about a 5-6 hour drive from Vegas . The much closer Grand Canyon West is a tourist trap run by the Native American tribe who's territory it's in. Basically, it's like expecting steak and getting an overpriced burger.
