Locals would not transfer from Keio to JR for that. From Shibuya, just get the Keio line (also not a subway). Mitaka is accessed easily by the Chuo local (yellow train) or Chuo Rapid (orange). From Shibuya you would ride the tree stops to Shinjuku and then transfer. It will not be on the subway map because it is JR and not a subway line.
